Bio-Rad has released technical note 5720, which describes a study identifying the optimal conditions for electroporating IMR-32 and SK-N-SH neuroblastoma cell lines with plasmid DNA or siRNA.
The study shows that good conditions for electroporating these cells are a 20ms square-wave pulse of 200V and 2,000uF.
Using a higher cell density yielded results that were just as good as those obtained with cells at lower densities, demonstrating that cell concentration is another important variable to consider when performing electroporations.
Further optimisation of cell density and plasmid DNA and siRNA concentrations can provide even better results.
