These new kits are designed to reduce preparation time for plasmid purification by nearly 70%, while yielding up to 100% more transfection-grade plasmid DNA than any other products on the market
The new Macherey-Nagel NucleoBond Xtra Midi/Maxi plasmid purification kits are now available through Thermo Fisher Scientific, Epsom, UK.
New plasmid purification kits deliver increased yields of transfection-grade plasmid DNA, including endotoxin-free preparations, while saving time in the laboratory.
NucleoBond Xtra is the first of a new generation of anion exchangers whose innovative column design allows clearing of the lysate to be accomplished in parallel with loading onto the column.
To ensure fast filtration, the column filter has a large structured surface which is additionally designed to minimise the risk of clogging.
High column flow rates are also achieved by the combination of a large column diameter with a low silica resin bed.
Further savings in preparation time result from the inclusion of NucleoBond Finalizer in the 'Plus' versions of both kits to speed up the rate of DNA precipitation.
The NucleoBond Xtra range now includes NucleoBond Xtra EF products for faster and higher yields of endotoxin-free plasmid DNA.
In tests, these kits have demonstrated excellent efficiency in providing endotoxin-free preparations (endotoxin levels using the standard or Plus versions of the NucleoBond Xtra Midi EF and Maxi EF kits are consistently <0.01EU/ug).
Typical plasmid yields from NucleoBond Xtra are >250ug (yields up to 540ug have been seen) for the Midi kit and >1000ug from the Maxi version.
These high yields are achieved through use of an improved silica material with high DNA binding capacity, together with optimised buffer compositions.
"In studies, NucleoBond Xtra has been proven to perform better than silica-membrane based kits in terms of both yield and preparation time, while also delivering superior purity of plasmid DNA due to use of anion-exchange technology", said Thermo Fisher Scientific's product life cycle manager, Valentina Janusic.
"Use of a desalting tool included in the NucleoBond Xtra Plus Midi and Maxi kits reduces the prep time down to 45 minutes, while providing recoveries up to 90%, depending on elution volume."
