High purity and high yield of genomic DNA from whole human blood via a two-stage protocol to first isolate and purify white blood cells, then bind DNA and remove contaminants
Thermo Labsystems has strengthened its KingFisher family with a novel automated method of isolating genomic DNA from human whole blood.
Designed for use in conjunction with the KingFisher mL instrument, the KingFisher Blood DNA kit employs a rapid two-step procedure, resulting in high yields of very pure genomic DNA.
The novel two-stage protocol ensures that only those blood cells containing DNA are isolated for purification.
The first step utilises Pan Leucocyte Dynabeads to isolate the white blood cells.
In this way, all impurities and unwanted material, such as red blood cells, are left behind.
The purified white blood cells are then washed, lysed and mixed with DNA-binding Dynabeads which bind the genomic DNA.
Subsequent washing steps remove unwanted cell debris and other contaminants, resulting in an average yield of 4-10µg of highly purified DNA from 200µl of blood.
Used in conjunction with the KingFisher mL, the kit offers two different protocol options.
DNA can be eluted either into a KingFisher mL tube, or into microcentrifuge tubes using a separate microtube tray.
This gives users the advantage of having the DNA ready for further analysis or storage immediately after purification.
The kit has been evaluated in Germany, UK, and USA, and is suitable for a wide range of applications, from mutation screening and SNP analysis to HLA or tissue typing.
Combining the new kit with the KingFisher mL instrument gives users a high level of automation, reducing the need for manual handling of potentially hazardous blood samples and increasing productivity.
