Anachem has introduced the DNAreleasy cell lysing reagent, which requires just a 10-minute incubation on a thermal cycler before the DNA is ready for use directly in PCR.
DNAreleasy has been used to lyse a wide range of cells including: human buffy coat, human fibroblasts tissue culture cells, human sperm, mouse and rat tissue culture cells, mouse tail, various plants, Drosophila, and others such as yeast, Gram positive bacteria, Gram negative bacteria, viruses and many more.
Using DNAreleasy requires the user to mix cells with 20ul of the reagent and overlay with mineral oil if necessary (for thermal cyclers without a heated lid).
Place this in a thermal cycler and treat at 75C for five minutes followed by 96C for two minutes before holding at 20C.
After the lysis, a part or all of the DNAreleasy lysate can be used directly in PCR.
DNAreleasy can make up to 40 per cent of most PCR mixes and can be stored at -20C for future use.
