Scientists at the Laboratory of Anthony L.
Fink (Department of Chemistry and Biology, University of California Santa Cruz) have demonstrated that Wyatt Technology's DynaPro Dynamic Light Scattering system is an effective alternative tool to in vitro experiments for following the protein unfolding process.
Protein folding and unfolding has historically been monitored via in vitro experiments, where the information obtained is directly related to the function and behaviour of proteins in vivo.
The aim of this experiment was to follow the unfolding process by another independent method - Dynamic Light Scattering.
Wyatt Technology's DynaPro Dynamic Light Scattering system is the industry standard in the protein crystallisation market.
The system offers the highest sensitivity and lowest sample volumes for protein crystallisation by DLS.
It comes with a unique range of features including a control module with the ability to collect and analyse data; and a read module, which comes as either a temperature-controlled scintillation vial reader for single measurements, or a fully automated plate reader with the capability to make measurements on 96 or 384 or 1536 well plates.
Other features include a stand-alone self-dessicating temperature chamber for low temperature conditions, and the ability to integrate seamlessly with Wyatt Technology's online accessories.
For this particular application, the Dynamic Light Scattering measurements were found to be capable of monitoring the dissociation of an oligomeric protein and unfolding of monomers simultaneously using chaotropic agents.
