The system's tolerance to stringent washing means background signals are reduced allowing the detection of small changes in protein binding patterns using minimal numbers of cells.
Proteins of interest are expressed as Halotag fusion proteins and cross-linked to their DNA binding sites with formaldehyde.
The Halotag protein allows immobilisation of this DNA:Protein complex to Halolink resin while the sample is washed to remove non-specific DNA and proteins.
The cross-linking is then reversed to release the target DNA for analysis.
The introduction of the Halochip system is a further addition to the range of assays that are facilitated using Promega's Halotag technology.
