Software package enables high quality analysis and excision from 1D polyacrylamide gels
A new software package is now available from Genetix for use with its GelPix system, which enables high quality analysis and excision from 1D polyacrylamide gels.
The GelPix system is a high-throughput protein spot excision system designed to accurately image, analyse and excise protein spots from self and pre-cast 1D and 2D gels.
The gels are imaged using a high-resolution on-board camera.
This image is then analysed by the integrated software, which generates a user defined protein target excision list.
The selected proteins are then excised using a novel patented eight-channel excision head, which allows for high-throughput, automated operation.
Using the QSoft Excision 1D package, the GelPix fully integrates Phoretix 1D software from Nonlinear Dynamics, which automatically detects lanes on the 1D gel and then locates the centre of each protein band.
The user can manually delete or add bands.
A CSV format picklist containing information on the selected samples including lane and band number, band intensity and molecular weight is then exported to the GelPix.
QSoft Excision 1D software offers a wide selection of excision options.
These include a single pick from the centre of each band or multiple picks from each band. Facility to excise overlapping plugs enables the excision of entire bands maximising the amount of protein sample for downstream processing.
As an alternative excision protocol, the 'click and pick' function, bypasses automatic spot detection and is the fastest way to select and excise specific bands.
Using this protocol provides the added convenience of being able to click and drag individual spot targets so that curved bands can be completely excised.
The analysis software also allows you to excise from gels that have been imaged remotely using the triangulation function.
The triangulation process is performed at the gel analysis stage and also allows excision of single or multiple picks from designated bands.
After excision the gel plugs are dispensed into 96-well microplates. The system offers the option to dispense all plugs from one band into a single well or to dispense plugs from one band into sequential wells.
The QSoft Excision 1D software package comes complete with a data tracking system.
