Fluorescence quench assay kits allow high throughput screening users to combine sensitive time resolution with signal increase measurement in enzyme degradation assays
PerkinElmer Life and Analytical Sciences, a provider of drug discovery, life science research, and analytical solutions, has launched TruPoint assay kits, a powerful solution for enzyme degradation assays for the drug discovery market.
Using PerkinElmer proprietary fluorescent SignalClimb technology, TruPoint assay kits provide superior sensitivity, precision, and reduce false positives, thus providing higher quality leads to pharmaceutical and biotechnology researchers conducting high throughput screening (HTS) in search of novel new drugs. Degradation enzymes, such as proteases and helicases, are a broadly studied class of therapeutic targets involved in many disease states, such as viral diseases and chronic inflammation. The TruPoint line kicks-off with three products: TruPoint Caspase kits for measuring caspase activity from apoptotic cell lysates and purified enzymes; TruPoint Helicase assay kits for measuring helicase activity; and TruPoint Labelling Service, which provides custom-made substrates created from peptide or oligonucleotide sequences provided to PerkinElmer by customers.
All of the TruPoint products were developed based upon PerkinElmer's SignalClimb technology.
This time-resolved fluorescence technology is able to measure enzyme activity through a signal increasing readout, as opposed to the traditional signal decreasing output.
This provides increased sensitivity, enabling measurement of low enzyme activities with high signal-to-background ratios, and little or no interference from background noise or coloured compounds.
Additionally, researchers find the ability to report out increasing enzyme activity with an increasing signal more relevant than decreasing signal assays.
