Purification of high quality plasmid DNA in just 30 minutes - claimed to be far quicker than with any other currently available system - is possible following the launch of this midiprep system
The PureYield plasmid midiprep system from Promega provides purification of high quality plasmid DNA in just 30 minutes. Furthermore, purified DNA can be used directly in downstream applications without the need for an alcohol precipitation step, since elution takes place in a small volume of water.
In addition to the improvement in speed, DNA yield has also been enhanced. Typically, 100-200ug of plasmid DNA can be obtained from 50ml of bacterial culture.
Furthermore, large culture volumes can be processed using an alternative lysate clearing protocol to produce yields of up to 400ug.
In use, the procedure is simply to add sample, wash and then elute.
The key is the PureYield membrane, a proprietary silica-membrane technology presented in a convenient dual-column format: an initial clearing column and a second, binding column.
When used with a vacuum manifold, both columns can be nested allowing clearing and binding in a single step.
An alternative, centrifuge-based protocol can also be used to run the columns sequentially.
Following binding, an endotoxin removal wash reduces contaminants (including endotoxin, protein and RNA giving high purity DNA, suitable for transfection, in vitro transcription/translation and fluorescent DNA sequencing.
