Simultaneous reactions are carried out in nine independent 96-well reactor stations.
The reactor stations are disposable micro plates, pre-filled with amide resin for the synthesis of 96 individual amide peptides.
To eliminate cross contamination of the building blocks (amino acids) a revolutionary dispensing technology has been developed: 'synthesis pens', reservoirs filled with the building blocks, which deliver a defined volume into the reactors.
The robotic arm picks up the pens, moves them to the reactor position and releases a defined volume into the reactor well.
There is an individual pen for each building block.
This speeds up synthesis, as no washes or flushes of tips or supply lines are needed.
Cross contamination is totally excluded.
For de-protection, washing and cleavage solvents or reagents are delivered by dedicated syringe pumps and distributed by a manifold.
At the end of the synthesis run, PepSy delivers cleaved and de-protected peptides amides: 864 10mer peptides of 1mg each per day! The software enables the user to create a peptide library very easily.
Peptide and protein sequences can be automatically imported, alanine-walks, substitution analyses and epitope mappings can be executed.
