Thermo Fisher Scientific has introduced its Thermo Scientific Extensor Long PCR Enzyme Mix for the amplification of long DNA fragments.
The Mix offers up to six times the fidelity and double the yield of regular Taq DNA polymerase in the amplification of long DNA fragments.
Thermo Fisher Scientific said this makes it ideal for applications such as cloning and mutagenesis or for the amplification of difficult and GC-rich fragments.
The blend contains ThermoPrime Taq DNA Polymerase and a proprietary, thermostable proofreading enzyme which act synergistically to ensure accurate and reliable amplification of DNA templates up to and in excess of 20kb.
The Extensor Enzyme Mix is available with two different buffers - one to optimise the amplification of fragments up to 12kb and the other for amplicons longer than 12kb or problematic PCR.
Each combination is also available pre-blended in a master mix format to simplify PCR set-up and reduce the risks of contamination or reagent handling errors.
These master mixes can be stored at four degrees Celsius, eliminating the need for freeze/thaw cycles that may reduce the activity of the enzymes.
Extensor Long PCR Master Mixes are also available with ReddyMix, which incorporates a unique red tracker dye and precipitant to allow direct loading of the PCR product onto gel for quick and easy post-PCR analysis.
