Bio-Rad's Silentfect lipid transfection reagent has been developed specifically for the delivery of siRNA to cultured mammalian cell lines.
Silentfect is described as being a novel, high-efficiency cationic lipid that delivers robust knockdown of endogenous targets with little cellular toxicity.
The very high efficiency of delivery means less lipid is required per transfection while still achieving gene knockdown of >90% for many cell lines.
In addition, the high affinity of Silentfect for siRNA means that as little as 5nM of siRNA can reduce target gene expression by as much as 90%.
The ability to achieve knockdown with low amounts of lipid and low concentrations of siRNA can also reduce the risk of off-target effects.
Optimal siRNA delivery and expression knockdown can be achieved with little toxicity in both serumcontaining and serum-free media.
The method is simple, requiring only three pipetting steps and a change of culture medium the day after transfection, says Bio-Rad.
