B-secretase (BACE1) is a key protease involved in the production of b-amyloid peptides found in extracellular amyloid plaques of patients with Alzheimer's disease (AD).
This enzyme has been implicated as an important target for anti-amyloid therapy of AD.
The recombinant human BACE1 contains the catalytic domain and a C-terminal His tag.
The protein was expressed as inclusion bodies in E.coli.
It was refolded and purified using ion exchange and IMAC chromatography.
Enzyme activity can been measured in fluorescence resonance energy transfer (FRET) substrate based assays (e.g SensoLyte 520 b-Secretase Assay Kit, cat# 71144) 100ng of B-secretase is sufficient for this FRET-based assay.
