The straightforward two-step principle of the Lumiprobe 24 tests comprises enrichment of the bacterial sample in a specific broth, followed by DNA/RNA hybridisation in the solid phase. Yielding results more rapidly than many other available methods, the entire process is complete within 24 hours as the reagents are 'ready to run', no RNA amplification is required, and there are just four basic handling steps - pipetting, washing, incubating and reading.
Following enrichment, the bacterial sample is placed into the microplate well or sample tube, the walls of which are coated with a specific probe that captures and fixes the target RNA released during bacterial cell lysis.
A second (labelled) oligonucleotide probe is hybridised to the RNA, and the bound hydrid is detected by an enzyme conjugate that interacts with the label on the probe.
The enzyme substrate is then added and detects the labelled bacterial RNA via a chemiluminescent reaction.
Definitive results are obtained in 24 hours without the need for confirmation, as specificity for Listeria and Salmonella is over 99%.
Offering versatile user options, the Lumiprobe 24 test is available in two formats - 50 tubes for individual and small volume test runs, and 96- or 192- well microplates for automation where large volumes and a range of tests are to be performed simultaneously.
