The newest version of Agilent's Multiple Affinity Removal System (Mars), the Human-14, removes 95 to 99 percent of the 14 most abundant proteins from human blood serum and plasma in one easy step
These high-abundance proteins account for 94 percent of the total protein mass, and removing them facilitates the discovery and identification of low-abundance proteins that may be biomarkers for a wide range of diseases.
Proteins associated with diseases are typically present in small amounts in human serum and may be difficult to isolate amid high-abundance proteins, such as albumin and immunoglobulin G.
By reducing by half the number of proteins masking potential biomarkers, the new Agilent Human-14 products help researchers delve more deeply into the human proteome and advance the progress of proteomic research.
The Mars Hu-14 joins Agilent's successful line of protein fractionation products.
The family includes: the Mars Human-7, Mars Human-6HC, Mars Human-6, and Mars Mouse-3 columns and spin cartridges.
"The Mars0-7 was excellent, and the Mars-14 is a major step forward for facilitating mining the plasma and serum proteome," said Ed Dratz of Montana State University.
"The Mars-14 is clearly improving the quality of our downstream analysis.
"We are incorporating the Mars-14 in our sample workflow, seeking early markers for type 2 diabetes".
Samples depleted with the Multiple Affinity Removal System are fully compatible with downstream tools without additional sample manipulation.
Dratz's sample workflow includes the Multiple Affinity Removal System, the Agilent 3100 Offgel fractionator, macroporous reverse phase C18 column and Agilent 1200 HPLC-Chip/MS system.
These tools expand the dynamic range of current LC/MS and electrophoretic analytical methods for analysing complex biological fluids and can be used to customise workflows including protein identification, characterisation or biomarker discovery.
The new Agilent Mars Hu-14 is available in both HPLC column and spin cartridge formats, facilitating protein removal through liquid chromatography or centrifugation techniques.
The new high-capacity products can be used for more than 200 runs, allowing immunodepletion of more than 8000 microlitres of serum or plasma using a single standard column format.
