Saving valuable time and money are the important benefits of Lyostat 2, the latest version of a compact freeze-drying microscope system, recently introduced by Biopharma Technology. Lyostat 2, which incorporates similar functions to the Lyostat 1 system, is invaluable for freeze drying cycle development, enabling quick and accurate identification of key parameters such as collapse temperatures.
The process begins with a small sample of product being loaded into a stage chamber under the microscope, which acts like a miniature freeze-dryer.
The sample is then frozen down to a pre-selected temperature and cooling rate, after which a vacuum is pulled in the chamber to begin the drying process.
The temperature of the stage can then be manipulated in order to identify the temperature at which the product will collapse or melt.
This process can be viewed either through the microscope, or via a computer using the camera and image capture program supplied with the system.
All software is now supplied as standard with Lyostat 2, allowing the user to control the freeze-drying remotely via a computer.
This includes displaying and recording temperature and vacuum readings, providing a live image of the sample to be viewed on the screen, as well as recording a gallery of images taken at selected time intervals.
One of the main differences between Lyostat 1 and 2 is the microscope.
With Lyostat 2 the use of a compound microscope is recommended, giving the option of using either transmitted or reflected light.
This enables the user to select the illumination best suited to view the sample and gives far greater magnification.
This means that the drying front and structure of the dried product can be seen much more clearly.
Polarisers and DIC (differential interference contrast), can also be used in conjunction with the microscope, enabling the identification of structural changes within the product.
The compound microscope for use with the Lyostat 2 can be purchased from Biopharma, or alternatively an existing compound microscope can be used if one is available.
Lyostat 2 has been designed to be compatible with most popular makes of micropscope, such as Olympus, Nikon, Leica, and Zeiss, making it a more cost- effective and flexible option. It is supplied fully validated against published eutectic or collapse temperatures for a range of standard solutions.
