For example, cloning of AT-rich genomic DNA is notoriously difficult, but using CloneSmart instead of conventional pUC based vectors has been shown to yield 25-30 times more stable clones.
Also, genomic libraries of small and large inserts can be generated with minimal effort.
The vectors in the CloneSmart kit are provided pre-cut and dephosphorylated, so researchers do not have to prepare their own vector. Furthermore, the CloneSmart system ensures there is virtually no background caused by empty vectors, so no screening for recombinants is necessary.
Therefore, all viable colonies can be picked for analysis, eliminating the error-prone manual or robotic discrimination between white, light blue and authentic blue colonies.
CloneSmart Blunt Cloning Kits incorporate pSmart vectors, which employ transcriptional terminators to minimise unintended transcription of the DNA inserts.
pSmart vectors do not use a promoter or indicator gene, so transcription through the insert is eliminated.
This lack of transcription greatly decreases the bias against cloning strong promoter regions, sequences that encode toxic peptides, or other unstable elements. These products are available from Cambridge BioScience in the UK and Ireland only.
