Designed to eliminate difficulties inherent in cloning, when tackling previously unclonable DNA which may result in sequence stacking, clone gaps or inability to create genomic libraries
Cambridge BioScience has introduced E cloni 10G chemically competent cells for use with the CloneSmart system from Lucigen Corporation.
E cloni 10G chemically competent cells have a transformation efficiency of >3x108cfu/ug, and can directly replace DH5alpha and DH10B cells.
CloneSmart was designed to eliminate many of the difficulties inherent in cloning, especially when tackling previously unclonable DNA which often resulted in sequence stacking, clone gaps or a complete inability to create genomic libraries.
The CloneSmart blunt cloning kits incorporate Psmart vectors which use transcriptional terminators to minimise unintended transcription out of DNA inserts.
The vectors do not use a promoter or indicator genes so transcription through the insert is eliminated.
This lack of transcription greatly decreases the bias against cloning strong promoter regions, sequences that encode toxic peptides, or other unstable elements.
99.5% of randomly picked clones contain insert DNA removing the need for blue/white screening for recombinants. These products are available from Cambridge BioScience in the UK and Ireland only.
