Rapid small-scale purification of GST-tagged proteins directly from cell lysates is now possible with the new MagneGST protein purification system from Promega.
Offering the flexibility of automated or manual methods, proteins can be purified to high yield with little or no non-specific background in under two hours.
Protocols can be adapted to meet specific needs, from 1-96 samples.
Similarly, volumes can be adjusted to suit the amount of material to be purified, from 1ml to 50ml.
Binding capacity for the MagneGST particles is 5-8mg protein per 1ml packed resin.
As the new system uses immobilised glutathione paramagnetic particles (MagneGST particles), there is no need for a centrifugation step to clarify the cell lysate.
This means it is readily adaptable for automation on suitable robotic platforms, such as the Biomek 2000 or Biomek FX.
Alternatively, a manual method with a variety of magnetic stands can be used.
This magnetic approach also avoids the sample loss often experienced using non-magnetic systems which are time-consuming and labour-intensive.
