A new 150x, 1.45NA oil objective offers excellent light transmission for total internal reflectance fluorescence (Tirf) research microscopy over an unprecedented range of the spectrum from the ultraviolet (UV) to the infrared (IR), says Olympus America.
The high-NA Olympus U Apochromat 150X Tirf objective enables easy Tirf observation using a standard coverglass and standard immersion oil.
Olympus says it offers the most comprehensive line of Tirf optics for advanced research.
The objective is the only one currently available that offers both high UV transmission down to 330nm and a very high numerical aperture, making it possible to perform Tirf microscopy and UV applications such as un-caging and/or photo-activation with just one lens.
In addition, the objective offers IR transmission well beyond 1000nm, making it well suited to IR fluorescence imaging and IR laser applications, and opening up the potential for live-cell microscopy and future applications in the growing IR microscopy field, sayus the company.
The high magnification of the objective enhances small specimen structures on the CCD camera, thus overcoming the resolution limit imposed by the CCD's pixel size, an advantage because it does not add either the light loss or potential for aberrations that additional magnification optics can cause.
In addition, due to its small field of view, the optic's illumination light is concentrated onto a very small area of the specimen, offering added illumination intensity that makes the objective a good choice for single molecule fluorescence applications.
The 150x NA 1.45 objective offers evanescent wave penetration characteristics similar to the existing Olympus 60x NA 1.45 Tirf objective.
It has a working distance of 0.10, a field number of 22, and uses a correction collar to compensate for coverglass thickness and temperature.
